How to generate quality nuclei suspensions from fresh, frozen, and FFPE tissues
The purpose of this seminar is to share with UF researchers, the Miltenyi tools available to generate quality single nuclei suspensions from tissues to support their downstream assays. The workflows and reagents introduced will support researchers to generate single nuclei suspensions from fresh, frozen, OCT-embedded, and even FFPE tissues. The latter, allowing labs to draw new data from archived patient samples. The workflow includes a MACS cell separation-based cleanup step, isolating nuclei away from debris to generate clean suspensions, acceptable for further downstream FACS sorting, or sequencing applications. This workflow is not species-specific and can even be used with plant samples.
The gentleMACS Octo with Heaters instrument is a key component to the workflows being presented, which is an instrument available for use in the ICBR Flow Cytometry Core, along with MACS Magnets for the nuclei cleanup steps.
